After completing his studies, Rami began working as a journalist at The Munsif Daily newspaper, where he regularly interacted with film personalities.

One day, Rami Reddy went to interview the renowned Telugu director Kodi Ramakrishna. Impressed by Rami's presence, Kodi Ramakrishna offered him a negative role in his upcoming film, Ankusam (1990). Rami accepted the offer, marking his debut in cinema. Ankusam became a blockbuster hit, launching Rami's career as a villain. He later reprised his role in the Hindi remake titled Pratibandh (1990), starring Chiranjeevi. The Hindi version also achieved success at the box office, and Rami's portrayal of Spot Nana garnered praise, making him one of the popular villains of that time.

His performances in movies such as Gunda (1998), Khuddaar (1994), Shapath (1997), and Waqt Humara Hai (1994) were well-received. Particularly, his portrayal of Colonel Chikara in the Akshay Kumar and Suniel Shetty-starrer Waqt Humara Hai became one of his most memorable roles.

During his career, Rami Reddy appeared in over 200 films across multiple languages, solidifying his presence in Bollywood during the 1990s and 2000s. His extensive filmography includes notable movies such as Andolan (1995), Dilwale (1994), Angrakshak (1995), Elaan (1994), Anaganaga Oka Roju (1997), Thulli Thirintha Kaalam (1998), Mrugaraju (2001), among many others.
